AstraZeneca has received approval from the US Food and Drug Administration for its anti-ulcer drug rosuvastatin calcium (Crestor), which is used to treat high blood levels of cholesterol, particularly at high doses. This approval is part of a large-scale phase III clinical trial to determine whether Crestor could be of benefit to the American public.
“Our study has demonstrated the cardiovascular benefits of rosuvastatin calcium compared to other statins,” said Steven Nissen, MD, of the School of Medicine, The Ohio State University School of Medicine.
Rosuvastatin calcium (Crestor) is an oral version of the statin cholesterol-lowering drug statins HMG-CoA and Meridia. Crestor has been used in the treatment of various lipid disorders such as atherosclerosis, hypercholesterolemia, and other cardiovascular diseases.
In the trial, Crestor was well-tolerated with a median plasma lipid levels of about 1.5 mmol/L (IQW) on day one of the study and a mean plasma lipid level of about 2.0 mmol/L (IQW) on day two of the study.
The trial also has a short duration of effectiveness, with an average duration of 3 months. The drug was well-tolerated with a median plasma lipid level of about 1.8 mmol/L (IQW) and a mean plasma cholesterol level of about 3.1 mmol/L (IQW).
The trial was stopped early after the study had shown a safety profile similar to Crestor. The drug is not expected to have a substantial effect on the incidence of heart disease, stroke, dementia, or other serious complications.
About 10,000 patients were treated at AstraZeneca. Patients were followed for up to three years.
The study was supported by the National Institutes of Health (NIDDK-117320-00), and by the National Heart, Lung and Blood Institute (NICHD-120989). The National Institute of Diabetes and Digestive and Kidney Diseases also supported the study.
“This trial demonstrates our ability to use this innovative approach to evaluate the cardiovascular benefits of statins for patients with high cholesterol and to determine whether Crestor could be of benefit to them,” said Michael T. Dolan, MD, Professor and Chair of the Obesity and Cardiovascular Sciences at the Cleveland Clinic. Dolan is the first to submit the trial results to FDA for approval.
